본 연구는 선천성알코올노출시후세대에미치는소아뇌발달장애의행동약리학적평가법을개발하고다세대전이성여부및그기전을후생유전학적연구를통하여확립함을 목표로 하였음. 상기 최종목표 달성을 위해
1) 인간의과도한음주상황과유사한임신중알코올섭취동물모델의제조
2) 태어난 실험동물의뇌발달장애를ADHD증상인과잉행동,충동성및주의력결핍을기준으로파악할수있는최적행동약리평가법개발
3) 개발된행동약리학적기법을적용하여알코올의 태내노출에의한태아의행동약리학적변동이F1동물교배시F2동물에세대간전이되는지의여부규명
4) 알코올섭취기왕력이두번째임신태아의ADHD증상에미치는영향규명
5) 태아알코올증후군에의한ADHD의세대간전이기전을후생유전학적차원에서규명하고자 하였음
- 인간의과도한음주상황과최대한유사한임신중알콜섭취동물모델의제조를 위해 경구반복투여에 의한 영향을 규명하였으며 임신 전 기간에 걸친 투여 보다는 임신 전기에만 알코올을 투여하는 것이 보다 효과적인 동물모델로 사용될 수 있음을 규명하였으며 흰쥐의 임신 6-15일 중에 1-6g/kg의 알코올을 투여할 경우 과도한신체적기형을유도하지않는조건에서신경학적증상을나타냄을 확인하였음.
-태어난실험동물의뇌발달장애를ADHD증상인과잉행동,충동성및주의력결핍을기준으로파악할수있는최적행동 약리 평가법을개발하여 Openfield,Foot-shock drinking paradigm,Y-Maze등의다양한행동약리학적검색기법을 적용하여 알코올이 과잉행동과 충동성 및 집중력 저해를 일으킴을 확인하였음. 이는 SHR등 다른 ADHD 모델 시스템에서 나타나는 양상과 유사하였음. 이러한 ADHD 행동 양상은 동물의 정상적 성장 패턴을 고려할 때 알코올에 의한 양육저해로 인해 야기되지 않음을 확인하였음.
-개발된행동약리학적기법을적용하여알코올의태내노출에의한태아의행동약리학적변동이F1동물교배시F2동물에세대간전이되는지의여부규명한 결과 ADHD 행동이 2세대에도 전이됨을 확인하였음. 한편 알코올 섭취의 기왕력이 있는 어미가 두 번째 임신을 하여 출산한 새끼의 경우에도 ADHD 증상이 나타남을 확인하여 임신 중 뿐 아니라 임신 전에도 과도한 알코올 섭취가 새끼에서 신경행동학적 변동을 일으킬 가능성을 확인하였음.
- 태아알코올증후군에의한ADHD증상과 이의 세대간전이기전을 규명하기 위해 노르에피네프린과 도파민, 세로토닌 수송체 등의 발현 양상을 검색한 결과 흰쥐의 forebrain에서 노르에피네프린 수송체의 발현이 증가하고 도파민 수송체는 감소함을 Western blot과 immunohistochemistry법에 의해 확인하였음.
- 이러한 변이가 임신 후에 유발되므로 후생유전학적 변이가 관여할 가능성을 규명하고자 노르에피네프린 수송체와 도파민 수송체 유전자의 methylation 변동을 methylationspecificPCR기법을 적용하여 분석한 결과 노르에피네프린 수송체의 methylation에는 큰 변동이 없으나 도파민 수송체 promoter의 methylation은 증가하는 경향을 나타냄을 확인하였음. 또한 methylation된 DNA에 결합하는 MeCP2의 발현이 증가되었음. 이러한 변동이 실제 도파민 수송체 발현 변동과 ADHD증상 발현에 영향을 미치는 지의 여부는 추후 더 많은 연구가 필요할 것으로 사료됨.

결론적으로, 흰쥐에서 임신 중 음주 혹은 임신 전 과도한 알코올 섭취는 후세대에서 2세대까지 전이되는 ADHD 증상을 유발하였음. 이러한 변이는 카테콜아민성 신경계의 변동에 의해 유발될 가능성과 이 기전에 후생유전학적 기전이 관여할 가능성을 확인하였으며 이는 알코올 위해관리 및 모자 보건에 중요한 연구결과로 사료됨.

6. 영문초록
The purpose of this study was to determine whether the exposure to EtOH in utero results in a series of physical and mental defects focusing on ADHD phenotypes such as hyperactivity, inattention and impulsivity. In addition, we tried to identify whether the prenatal alcohol-induced ADHD phenotype is transmitted to second generation of the offsprings and to investigate the mechanism mediating these effects focusing on epigenetic mechanism such as alterations in methylation of key neurotransmitter transporter systems. In this study, we investigated the effect of prenatal exposure to EtOH on impulsive behavior of young Sprague- Dawley rats using delay aversion apparatus designed to punish thirsty rats by electro-foot shock whenever they try to cross the shock area to drink fresh water. Oral administration of 25% EtOH to pregnant rats did not change the number and weight of the offspring but increased spontaneous exploratory activity in an open field test. Prolonging the treatment period to 14 days inhibited the locomotor activity probably due to either increased anxiety or defects in motor function. Despite the foot shock, young rats from EtOH treated mother tend to take risk to cross the electroshock area to drink water. Both the number of drink trial and duration of staying at the shock and water area were significantly increased in prenatally EtOH-exposed rats. Offsprings from the EtOH treated mother also showed decreased spontaneous alteration in Y-Maze task suggesting these animals suffer attention deficits. Interestingly, the ADHD phenotype also observed in the second litter of previously EtOH-treated mother rats suggesting that maternal alcohol exposure may affect ADHD phenotype not only during pregnancy but also before pregnancy. In addition, the ADHD phenotype was transmitted to the second generation of prenatally alcohol exposed rats. We next investigated the catecholamine transporter expression and prenatal EtOH exposure increased norepinephrine transporter expression and decreased dopamine transporter expression in Western blot and immunohistochemistry. Methylation specific PCR showing increased methylation of DAT gene promoter as well as increase MeCP2 expression suggest that epigenetic mechanism underlies the down-regulation of DAT transporter expression. In summary, these results suggest that prenatal alcohol-induced ADHD is transmitted multi-generationally by mechanism involving epigenetic alterations in catecholaminergic system in rat. The exact mechanism and the extent of milti-generation transmitability should be investigated further in the future study.
